SoluLinK Crosslinkers: HyNic, 4FB and other Crosslinking Kits


The SoluLinK crosslinking method is a unique, simple method for linking two biomolecules much more effectively than previously possible with standard crosslinkers. This method requires a three-step reaction:

  1. Activation of Biomolecule #1 withLinker A (HyNic)
  2. Activation of Biomolecule #2 with Linker B (4FB)
  3. Mixing the two activated biomolecules to form a conjugate

This three step process is better than past methods as it does not form homo-polymers and allows for more controllable formation of the desired conjugate. The Hynic and the 4FB linkers are available in various lengths and may be chosen to react with either amine or thiol groups on biomolecules. To get started, simply order the kit (S-HyNic Conjugation Kit) or their two individual reagents (S-HyNic and S-4FB).

These crosslinkers are the most efficient and quantitative linkers available on the market and produce quality, easily reproducible conjugates for better performance in your assays.


All conjugations utilizing the SoluLink Crosslinkers require linkers from both the Linker A and
Linker B Groups.

Sulfo-S-HyNic Crosslinker

Sulfo-S-HyNic is the water soluble version of the standard S-HyNic linker. It is used to HyNic activate amine containing biomolecules for conjugation to 4FB containing biomolecules or surfaces. The Sulfo version of the S-HyNic is typically used for proteins and surfaces sensitive to organic solvents.

SHTH Crosslinker

SHTH is used to activate amine containing biomolecules for conjugation to 4FB containing biomolecules or surface. The SHTH forms a non-aromatic bond with S-4FB and therefore forms a less stable hydrazone bond than does the S-HyNic with 4FB. SHTH is most often used to activate oligos for conjugation to 4FB-activated biomolecules to prevent dimerization of the oligo. An additional feature of SHTH is that it is more stable on the activated biomolecule than the HyNic for long-term storage before conjugation with its 4FB compliment.

C6-S-4FB has a 6 carbon chain for extended linker length. C6-S-4FB is similar to S-4FB in its reactivity and long-term stability. The hydrophobic 6-chain carbon linker does make proteins more sensitive to precipitation and should not be used to modify proteins with known solubility issues.
